Sepsis

Sepsis is a serious medical condition characterised by dysregulated systemic inflammatory responses followed by immuno-suppression.  It is a common syndrome that is responsible for killing thousands of patients each year but is still poorly understood.

Axis Bio offer the cecal ligation and puncture (CLP) and LPS-induced models to study pathogenic mechanisms and identify potential therapeutic agents to combat sepsis.

  • Cecum Ligation and Puncture (CLP)-Induced Sepsis model: This is a widely used model which closely resembles the progression and characteristics of human sepsis. CLP-induced rodents display pathologies that are seen clinically with humans, including hypothermia, tachycardia and tachypnoea. This is the most clinically relevant research model and can be adapted to test a range of severities (initially presents as polymicrobial peritonitis, translocation of bacteria into the bloodstream, septic shock, multi-organ disfunction and finally death). Read-outs include clinical observations, PD blood collections and histological evaluation.
  • LPS-Induced model: LPS administration induces system inflammation that mimics the early clinical features of sepsis. This model has a very rapid onset of disease and the associated increase in cytokines is very high compared to that seen in human clinical disease and is transient in nature. Although this model is useful, particularly for assessing PD response of cytokines, the acute nature should be noted.
