Rheumatoid Arthritis

Inflammation Models – Adjuvant induced RA model - rat

The rat AA model has been widely used in preclinical studies to design and test new therapeutics. The major advantage of this model is the rapid onset (10 days aDer immunization) of disease and easily measureable progression.

The model system at Axis Bioservices is immunization of Lewis rats with complete Freund’s adjuvant on day 0 . Disease is evident from day 10 and study continues for a further 5 days. Treatment can be commenced at any stage to demonstrate a prophylactic or therapeutic effect.

Dosing opIons/Time frames

While all our protocols are completely flexible we have found that it can help with study design if we provide some timeframe advice.

ProphylacIc Dosing: Begin dosing on day 0 (date of initial immunization) and continue until necropsy (day 15).

TherapeuIc: Begin on day 8 and dose for 7 days until necropsy (day 15).

Inflammation Models – Collagen induced RA model - mouse

CIA is one of the most extensively studies models of rheumatoid arthritis and has been widely used to identify pathogenic mechanisms of autoimmunity, as well as to design and test new therapeu&cs. The major advantage of this model is that it has many features of human disease.

The model system at Axis Bioservices is immuniza&on of DBA/1 mice with CII emulsified in complete Freund’s adjuvant on day 0 followed by a boost on day 21 . Treatment can be commenced at any stage to demonstrate a prophylactic or therapeutic effect.

ArthriIc Index Scoring

System is based on evidence of erythema and swelling.

0 = Normal

1 = One paw joint affected or minimal diffuse effects.

2 = Two paw joint affected or mild diffuse effects.

3 = Three paw joint affected or moderate diffuse effects.

4 = Four paw joints affected or marked diffuse effects.

5 = Erythema and severe swelling encompass the ankle foot and digits.

Dosing opIons/Time frames

Prophylactic Dosing: Begin dosing on day 0 (date of initial immunization) and continue until necropsy (day 35-45).

Therapeutic: When animal has an arthritic index of 1 dosing is commenced (day25-27), and continues for 14 days.
